Interactions between tumour cells and microenvironments may affect their growth and metastasis formation. Several studies have demonstrated the important role of cellular mediators in the progression of breast cancer cells. The effect of pro-inflammatory cytokines, such as IL-1, IL-6, TNF-α and IFN-g on the regulation of expression of chemokine receptors and autocrine-paracrine positive feed-back loops for releasing of inflammatory factors are the key steps in regulating trafficking and metastasis. Indeed, breast cancer cells express chemokine receptors in a non-random manner, and these observations pointed to several chemokine/ receptor pairs that control cell–cell migration.
This thesis gives some light to understand the interplay between tumour cells, stroma/inflammatory cells and inflammatory cytokines/chemokines and let us to propose additional levels of cross-talk between tumour cells and inflammatory mediators which are taking place in the surrounding microenvironment. In addition, a better understanding of the role played by the inflammatory components in breast cancer progression may also lead the way to the find of new therapeutic targets.
